AFM: a versatile tool in biophysics

چکیده
Here we review the applications of atomic force microscopy to the study of samples of biological origin. Emphasis is given to provide the reader with information on the broad range of different biophysical applications that, to date, such a technique can deal with. After recalling briefly the operating principles of an atomic force microscope, the broad field of bio-imaging applications is faced (DNA, DNA–protein interaction, proteins, lipid membranes, cells); thereafter, the use of the atomic force microscope to measure forces is introduced and force mapping on living cells is discussed. This section is followed by the description of the use of force curves in assessing single-molecule interand intramolecular interactions. A paragraph on the perspectives of the technique in biophysical applications concludes the paper. We hope that this review can help the reader in appreciating how atomic force microscopy contributes to the current explosive growth of nanobiosciences, where biology, chemistry and physics merge.

Demonstration of correlative atomic force and transmission electron microscopy using actin cytoskeleton
In this study, we present a new technique called correlative atomic force and transmission electron microscopy (correlative AFM/TEM) in which a targeted region of a sample can be observed under AFM and TEM. The ultimate goal of developing this new technique is to provide a technical platform to expand the fields of AFM application to complex biological systems such as cell extracts. Recent adva...
متن کاملAtomic force microscopy in biomaterials surface science.
Recent progress in surface science, nanotechnology and biophysics has cast new light on the correlation between the physicochemical properties of biomaterials and the resulting biological response. One experimental tool that promises to generate an increasingly more sophisticated knowledge of how proteins, cells and bacteria interact with nanostructured surfaces is the atomic force microscope (...
متن کاملMeasuring the elastic properties of living cells with atomic force microscopy indentation.
Atomic force microscopy (AFM) is a powerful and versatile tool for probing the mechanical properties of biological samples. This chapter describes the procedures for using AFM indentation to measure the elastic moduli of living cells. We include step-by-step instructions for cantilever calibration and data acquisition using a combined AFM/optical microscope system, as well as a detailed protoco...
متن کاملSample preparation procedures for biological atomic force microscopy.
Since the late 1980s, atomic force microscopy (AFM) has been increasingly used in biological sciences and it is now established as a versatile tool to address the structure, properties and functions of biological specimens. AFM is unique in that it provides three-dimensional images of biological structures, including biomolecules, lipid films, 2D protein crystals and cells, under physiological ...
متن کاملStructural and functional imaging with carbon nanotube AFM probes.
Atomic force microscopy (AFM) has great potential as a tool for structural biology, a field in which there is increasing demand to characterize larger and more complex biomolecular systems. However, the poorly characterized silicon and silicon nitride probe tips currently employed in AFM limit its biological applications. Carbon nanotubes represent ideal AFM tip materials due to their small dia...
